Our team, made of clinical and basic scientists working closely together, is recognized for its expertise in nutrition and epithelial inflammation. The general topic of our team is investigating inflammation in cystic fibrosis and inflammatory bowel disease.


Research overview

We are studying the mechanisms of action and regulation of nutritional factors in situations of inflammation and infection. Our hypotheses are that new nutritional components (mainly issued from breast milk) could influence:
(i) the immune system (gene expression) early in life and prevent and/or modulate inflammatory diseases later in life,
(ii) the course of inflammatory and/or infectious disease such as inflammatory bowel disease and cystic fibrosis.
Our aim is to understand at different periods of prenatal and postnatal development the mechanisms by which nutritional components can influence future health and prevent/treat inflammatory diseases.



Polymeric mucins form the matrix of mucus gels and are responsible of the biochemical and physical properties of the mucus. Mucins modulate the access, cell adherence, and colonization of the respiratory and gastro-intestinal tract of all vertebrates. We hypothesized that reinforcing the mucus barrier by nutrition or delivering recombinant mucin domain could limit or stop colonization and translocation of pathogens.



Inflammation and nutrition

The effects of maternal nutrition are well recognized. Both mendelian and non-mendelian (eg epigenetic) genetic factors influence gene expression and it is now clear that maternal nutritional status plays a major role in fetal and postnatal development and in health of the offspring and even later generations. In this context, nutritional status of the young infants likely affects their health. We aim to identify new alimentary compounds to prevent and/or modulate the immune and inflammatory systems and reinforce the mucosal barrier during lung and intestinal infectious diseases. The programming effects of new alimentary compounds during prenatal and early postnatal life will be also assessed on growing animals (intestinal development, immune function, nutritional status). Our results from clinical research in bronchopulmonary dysplasia and cystic fibrosis showed that inflammation has nutritional consequences potentially reversible notably by antibiotic treatment of the acute bronchopulmonary exacerbations by P. aeruginosa in cystic fibrosis. We showed that the long-term nutritional and respiratory consequences of bronchopulmonary dysplasia in young children were strongly influenced by the nutritional status during the first 2 years of life. In cystic fibrosis, pulmonary involvement is facilitated by early inflammation, gradually complicated by bronchopulmonary infections. In a mouse model of lung infection, we showed that long chain unsaturated fatty acids (PUFA) of the series n-3 prevent and/or reduce the alteration of lung function and improve prognosis. A clinical trial in cystic fibrosis patients is ongoing to assess the effect of n-3 PUFA on respiratory mucin expression.

Our team also develops clinical (intervention) research and epidemiological approach using our pediatric medical ward, as well as the pediatric clinical investigation center (set up in January 2011, one of the 3 clinical investigation centers specifically devoted to children in France), and a European network HELENA (Healthy Lifestyle in Europe by Nutrition in Adolescence, 6th PCRD- FOOD-CT-2005-007034). Recently, the impact of neonatal nutritional environment (ie birth weight and breast feeding) on nutritional status and inflammation at adolescence was investigated giving rise to more than 35 international peer publications.

The findings of this research will increase our knowledge of the health benefits of dietary compounds. Furthermore, this research program will provide nutritionally-based therapies in prevention or treatment of inflammatory diseases and may suggest candidate-compounds for infant formula.


Inflammation and the mucosal barrier

We are studying mucins of cystic fibrosis (CF) patients and in mouse models of CF with a specific focus on gel-forming mucins. Mucins are large and heavily O-glycosylated molecules that form long polymers. The two main mucins secreted in the lung are MUC5B and MUC5AC. Both MUC5B and MUC5AC contain multiple copies of a highly conserved domain found more than 25 times in a secreted mucin of the fish. A better understanding of the structure and the function of this domain that seems to be able to form non covalent bonds is essential to propose a new therapeutic approach in CF. Mucus plugs and mucin expression pattern in a mouse CF model are studied. Homemade knock-in mouse models for several mucin genes and recombinant molecules are investigated in this project. We designed also a transgenic mouse line that secretes into the lumen of its intestine a recombinant molecule eGFP-tagged which should improve the mucus gel barrier in experimental bacterial infections. A better knowledge of the first innate barrier of the intestinal tract may add to information about the interaction between intestinal microflora and enterocytes and will allow for a better understanding and treatment of inflammatory and infectious intestinal diseases.
